Abstract

Oral drug delivery system has numerous advantages compared to parenteral injection, however some peptide and protein drugs occur degradation by gastrointestinal enzyme when given orally. In this study, ovalbumin-alginate microsphere were prepared to protect ovalbumin from degradation and enhance the uptake of ovalbumin-alginate microsphere by lymphoid tissue in mice’s intestine. Ovalbumin-alginate microsphere were made by ionotropic gelation method with aerosolization technique using Na-alginate as polymer and CaCl2 as cross-linker. To increase microsphere’s stability during storage, microsphere formed were dried using Freeze-Dryer and maltodextrin 5% was added to protect microsphere during lyophilization. In vivo uptake study of microsphere in mice’s villi and Peyer’s patches at different time series were perfomed by using fluorescence microscope with rhodamine B as fluorochrome. The result, both ovalbumin-alginate microsphere and ovalbumin-alginate microsphere with maltodextrin 5% as lyoprotectant were able to be taken up by villi at 6 hours after given orally and taken up further by villi and Peyer’s Patches at 7,8,9,10 hours after given orally. Replicate samples are needed to be analyzed statistically.
