Abstract
Ezetimibe has been classified as Biopharmaceutics Classification System (BCS) class II category which generally has high permeability but poor solubility. This problem will cause low dissolution rate, absorption and bioavailability. The aim of the present study was to enhance dissolution rate of ezetimibe through solid dispersion which is developed in combination with surface adsorption techniques (surface solid dispersion). PEG 6000 was used as hydrophilic carrier and combination Avicel PH 101 with lactose monohydrate in 1:2 ratio was used as an adsorbent. Solid dispersion and surface solid dispersion were prepared by melthing method. Dissolution test and physical characterization were carried out to evaluate the formation of the both system. Dissolution of solid dispersion and surface solid dispersion was enhanced with increased PEG 6000 concentration compared to ezetimibe. Surface solid dispersion showed higher dissolution efficiency and dissolution rate than solid dispersion and pure drug. Characterization using X-ray diffraction showed that there was a decrease of ezetimibe’s crystalline peak intensity in the solid dispersion and surface solid dispersion compared to the ezetimibe. The result of differential thermal analysis indicated that the melting point of ezetimibe in the solid dispersion was disappear.
