Abstract
The purpose of this study is to determine synergistic in vivo effect on anticancer activity of Doxorubicin in combination with Diterpene Lactone Fraction of “Sambiloto” (Andrographis paniculata Nees) on fibrosarcoma in mice induced by benzo(a)pyrene. Fibrosarcoma cancer was induced by 0,6 mg/0,2 ml benzo(a)pyrene 0.3% in each male mice subcutaneously around the neck five times every two days. The mice were treated for about two month until the cancer grown. Then the mice divided to four different dose groups of treatment. First group as negative control, the mice were given CMC-Na 0.5% (15 days). Second group was given 594.80 mg/ kg body weight diterpene lactone fraction of “Sambiloto” by oral route (15 days). Third group was given 1,2 mg/kg body weight doxorubicin by i.p injection on the first day of treatment. The fourth group was given the combination of 594,80 mg/kg body weight diterpene lactone fraction of “Sambiloto” by oral route (15 days) and 0,6 mg/kg body weight doxorubicin by i.p injection on the first day of treatment. After treatment finished, the mice were sacrificed and dissected in order to separate the cancer tissue. The cancer tissue were weighted and processed into histopathology preparation slices stained with Hematoxylin-Eosin (HE). The synergistic effect was observed from cancer weight data, analized with one way ANOVA, and scoring of histopathology neoplastic changes analyzed with Kruskall-Wallis statistic method. In this study, based on histopathology results, it is concluded that the combination of Diterpene Lactone Fraction of “Sambiloto” (Andrographis paniculata Nees) at 594.80 mg/kg body weight of mice and Doxorubicin at 0,6 mg/kg body weight of mice has a synergistic in vivo effect on fibrosarcoma anticancer activity.
