Abstract
Background: Secretome or molecules secreted into the extracellular space obtained from stem cell culture media, obtained from Adipose mesenchymal stem cell (AMSC) has an advantageous effect for ocular surface regeneration. Purpose: To investigate the effect of Adipose Mesenchymal Stem Cell (AdMSC) Secretome in rabbit model severe dry eye disease, the clinical profile of TBUT, STT, corneal epithelial defects, cytokine pro-inflammatory IL-1 in ocular surface. Methods: We conduct Pre-test and Post-test experimental study of topical instillation of AdMSC secretome group, balanced salt solution group, and normal control group. Animal model was New Zealand white rabbit induced severe DED model using a combination technique of lacrimal gland resection and concanavalina injection. Rabbits were treated with adipose SPM secretomes or BSS, for seven days. Tear production by Schirmer tear test I, corneal fluorescein dye staining, inflammatory cell and cytokine pro-inflammatory IL-1 in ocular surface were analyzed Results: Topical instillation or AdMSC secretome successfully improved tear production by Schirmer tear test result, and lengthening TBUT time, reduced inflammatory cell and cytokine pro-inflammatory IL-1 in ocular surface Conclusion: The results of this study found that administration of SPM adipose secretome was improving clinical profile of TBUT, STT, and corneal epithelial defects, statistically significant. The mean value of IL-1 expression had a lower mean expression value in the Adipose SPM Secretome group than BSS group. Our result of topical instillation or AMSC secretome in animal model for DED has a great potential as a future cell-free based therapy for ocular surface.
