Abstract

Cancer is still the leading cause of death globally. Naringin is reported as a natural compound that has potential effect as anticancer. Naringin could be used as suppressor agent in the formation of making new cancer cell and as an inhibitory agent to stop carcinogenic that cause cancer initiation. Therefore, this study was conducted to analyze the anticancer effect of attaching folate on nanoparticles naringin (FA-NNPs) on T47D cells. FA-NNPs characteristics is tested using PSA (particle size analyzer), TEM (transmission electron microscopy) and FTIR (Fourier transform infrared). Cytotoxic activity is performed using the MTT test (3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium bromide), the migration cells are also analyzed, imunocytochemistry test was carried out by the expression of caspase-3, cell cycle tests to determine cell inhibition and the percentage of cells undergoing apoptosis using flowcytometry, detection of MMP-2 (matrix metalloproteinase-2) and TIMP-2 (tissue inhibitors of matrix metalloproteinase-2) genes to determine the inhibition of metastasis using real-time RT-PCR. This study showed that FA-NNPs had a particle size of 92.50 % below 100 nm. FA-NNPs have cytotoxic activity on T47D with an IC50 of 100 μg/mL, inhibited T47D cell migration and increased caspase-3 expression 67.80 %. The percentage of T47D cell apoptosis 92.50 % of cells experienced apoptosis. The cell cycle was inhibited in the G0/G1 phase. The result of the MMP-2 gene expression ratio is below normal cells while the TIMP-2 gene expresses 2.5 times more than normal cells. Based on the results of the study, FA-NNPs have been shown to inhibit the growth of T47D cells selectively through the folate receptor pathway on the surface of cancer cells to perform intrinsic apoptosis.
