Abstract
In recent years, the preparation, properties, and use of carbon nanodots (CNDs) derived from natural products gained great attention because natural products are the effectual starting materials to prepare CDs with typical physicochemical properties. This study is aimed to produce catechin-based carbon nanodots (Cat-CNDs) as staining agents for tumor cells. In this dissertation, Cat-CNDs were prepared from catechin isolated from Uncaria gambir (U. gambir) through a simple and facile pyrolysis method at 250C for 4 hours, which allowed it for carbonization. After that, solid black particles of Cat-CNDs were produced and further dissolved in NaOH. The resulting Cat-CNDs were characterized using significant analytical techniques. The results showed that unique physicochemical properties of Cat-CNDs such as photoluminescence with emission peak centered at 500 nm (ex =380 nm) and its average size distribution of 5 to 10 nm, and its cell viability of over 85%, the as-prepared Cat-CNDs were incredibly potential for staining targeted tumor cells. The Cat-CNDs also retained the biofunctions of catechin and were capable of being selected for staining specific tumor cells according to the spectroscopic analysis. Furthermore, the in vitro confocal microscopy analysis demonstrated their strong fluorescence which meant that the Cat-CNDs easily penetrated Hela cells via endocytosis. The CCK-8 assay analysis confirmed the high percentage of cell uptake viability due to its non-toxic behavior. The overall results showed that Cat-CNDs were dispersible and water-soluble, favorable hydrophilicity, excellent photostability, good colloidal stability, good biocompatibility, and non-toxicity. Therefore, this work highlights the as-prepared Cat-CNDs which may have great potential for an efficient staining agent of targeted tumor cells.
