Abstract

Alzheimer’s Disease (AD) is a progressive neurodegenerative disease that affect cognitive abilities, which is the most common cause of dementia. AD often linked to neurotransmitter deficiency, acetylcholine (ACh). Uses of cholinesterase inhibitors for AD treatment are common. Many researchs reported the main phytoconstituent responsible to acetylcholinesterase (AChE) inhibitory activity is alkaloids. Senna spectabilis has been known contains piperidine alkaloids such as cassine, spectaline, and 3-O-acetylspectaline. In the previous study it has been reported that the ethanol extract of the leaves of S. spectabilis inhibited AChE with IC50 value of 53,53 μg/mL. Investigation on the fractions showed that the ethyl acetate and n-butanol fractions of S. spectabilis extract gave strong inhibitory activity, with IC50 values of 10,77 μg/mL and 12,77 μg/mL, respectively. The aim of this study was to examine the AChE inhibitory activity of the n- butanol subfractions of S. spectabilis L. Vacuum Liquid Chromatography (VLC) was used as method for the fractionation to produce 12 subfractions. All subfractions showed ≥50% of inhibitory percentage after the AChE inhibitory activity assay using Ellman’s method. The IC 50 analysis was conducted on subfraction 3 and 5. Subfraction 3 gave IC50 value of 8,07 ± 0,85 μg/mL meanwhile subfraction 5 gave 29,58 ± 11,85 μg/mL. These results suggest that S. spectabilis has potency as acetylcholinesterase inhibitor.
