Abstract

Nicotine is a psychoactive compound from the tobacco plant and causes addiction. Nicotine addiction occurs because nicotine causes the activation of nicotinic acetylcholine receptors (nAChRs) on the release of dopamine by the mesocorticolimbic system. Increased production of reactive oxygen species (ROS) by nicotine and cigarette smoke extract (CSE) causes dopamine to increase and leads to nicotine addiction. Therefore, andrographolide, a natural antioxidant, is needed to reduce ROS and increase endogenous antioxidants so that the risk of nicotine addiction decreases. This research was carried out to analyze the effect of nicotine and CSE in causing a reward effect with the conditioned place preference (CPP) method. The experimental animals used were 5 male Mus musculus, Balb/c strains per group. The drugs injected were normal saline 1.0 mL/kg subcutaneous as control group, nicotine 0.5 mg/kg subcutaneous, CSE 1.0 mg/kg subcutaneous, and andrographolide 50 mg/kg intraperitoneal as pre-treatment. The result measured in this study is the reward effect through the CPP score using the biased design method. The results of post-conditioning tests showed that nicotine and CSE causes the increase of the reward effect significantly (p<0.001). While in the nicotine + andrographolide group (-5.3 ± 24.04) s and CSE + andrographolide group (-12.7 ± 17.96) s there was no increase of the reward effect. The results of this research show that andrographolide can lower the risk of nicotine addiction.
