Abstract

Ursolic acid is a triterpenoid compound that effectively inhibits tumor growth through apoptosis, inhibition of cell cycle, autophagy, and hepatoprotective. However, ursolic acid has poor solubility and penetration. Niosome has been reported to improve the bioavailability of drugs with low solubility. This study aimed to evaluate the in vivo biodistribution of chitosan coated ursolic acid niosomes in plasma and liver of mice. At first, the analytical method was optimized. Then, the plasma and liver were homogenized and prepared for ursolic acid determination. The HPLC optimum method involves the use of mobile phases consisting of acetonitrile : 0.5% acetic acid (90:10) for plasma analysis, and acetonitrile : 0.5% acetic acid (70:30) for liver organs. The samples were eluted through ODS Hypersil 150x4.6mm column at a flow rate of 1 mL/min in room temperature (22oC). The volume of injection was 20 μL and analyzed for ursolic acid level at the analytical wavelength of 210 nm. The result showed that chitosan coated niosomes had higher concentration of ursolic acid than niosomes without chitosan coated and suspension of ursolic acid in mice plasma. Meanwhile in the liver, the ursolic acid concentration in chitosan coated niosomes treated group showed not significantly different from that of niosomes without chitosan coating. The ursolic acid suspension showed the highest ursolic acid concentration in the liver. In conclusion, addition of chitosan on ursolic acid niosomes successfully increased the plasma level and did not significantly affect the liver level administered orally in mice induced with N-nitrosodiethylamine.
