Abstract

The present study was designed to investigate effect of cimetidin on orally ciprofloxacin metabolism in rabbit. Simple, rapid, reliable and sensitive ctrofluorometric methods were developed for the determination of ciprofioxacin. The methods depend on the chelation of the drug with molybdenum to produce uorescent chelates. At the optimum reaction conditions. ciprofloxacin-molybdenum helates showed excitation 279 om and emission 445 run. The effect of cimetidine on e pharmacokinetics of ciprofloxacin was assessed in four healthy rabbits ciprofioxacin (23 mg/kg body weight) was administered orally with cimetidine (37 mg/kg body weight). Oral cimetidine was administered on pre-treatment once a day for even consecutive days. Blood samples were drawn over at 0. 15.30.60.90. 120. 150, 80, 240, 300, 360. 480 minuts after administration. The apparent totall body clearanced the terminal elimination rate constant were calculated for each subject prior to and after cimetidine pre treatment. Total body clearance and the terminal elimination rate constant values for the combination treatment were stastical1y not different from the iprofioxacin treatments alone (clearance and the terminal elimination rate constant values prior to and after cimetidine pre treatment were 0,34 ± 0,35 and 0,18 ± 0,26 minutes; 59,39 ± 35,89 and 299,02 ± 294.85 minutes) respectively. The concomitant administration of ciprofioxacin and cimetidine compared with control increased but not significant.
