Abstract

The objective of this study was to get the specific protein of T. canis and to in estigate the dominant immunoglobulin that could be used as a biomarker in kit di gnostic to toxocariasis diagnosis through antibody test by indirect-ELISA technique. This study was done to isolate and characterize T. canis protein from e cretory-excretory (ES) material of 12 dormant by pure protein that was reactive with p Iyclonal antibody. !n the first step, 12 dormant were homogenized by sonication 3 30 seconds and identified by SOS-PAGE with silver staining. The second step, p tein was transferred to nitrocellulose membrane used polyclonal antibody of antidormant that was visualized through goat-anti rabbit conjugate and SCIP+NST st ining. The third step was decided the specific of protein fractions that was done b se on molecule weight (MW) and the specific protein was isolated by elution te hnique. The fourth was to evaluate the antigenicity, sensitivity and specificity of c de and pure protein by IgG-ELISA as response against T. canis infection toward m e and to study the profile IgG and the dominant of subclass IgG as response ag inst T. canis infection toward mice. The results of [he study that: 1) T. canis have more protein with variation of m lecular weight between 8 and 200 kOa; it also the specific and cross, reaction pr teins of T. cati and antibody against T. canis; 2) The specific proteins of T. cati that and recognized by the anti-To canis sera are as follow proteins of MW 168,120,80 and 70 kOa; 3) The specific subclass of Ig G in the anti-To canis mice sera were IgG2a; 4) Saed on the ELISA 00 values, pure protein Toxocara spp. have high antigenicity of se um anti-Toxocara spp., they have a lower antigenicity against other worms (A cylostoma spp. and D. ,caninum), with a value of 1000.4 sensitivity and specificity of 875%.
