Abstract

Background: Coronavirus disease2019(COVID-19)isaglobalhealthproblemthatcausesmillionsofdeaths worldwide. TheclinicalmanifestationofCOVID-19widelyvariesfromasymptomaticinfectiontoseverepneumonia and systemicinflammatorydisease.Itisthoughtthathostgeneticvariabilitymayaffectthehost’s responsetothe virus infectionandthuscauseseverityofthedisease.TheSARS-CoV-2virusrequiresinteractionwithitsreceptor complex inthehostcellsbeforeinfection.Thetransmembraneproteaseserine2(TMPRSS2)hasbeenidentifiedas one ofthekeymoleculesinvolvedinSARS-CoV-2virusreceptorbindingandcellinvasion.Therefore,inthisstudy, we investigatedthecorrelationbetweenageneticvariantwithinthehuman TMPRSS2 gene andCOVID-19severity and viralload. Results: We genotyped95patientswithCOVID-19hospitalisedinDrSoetomoGeneralHospitalandIndrapuraField Hospital (Surabaya,Indonesia)fortheTMPRSS2p.Val160Metpolymorphism.Polymorphismwasdetectedusinga TaqMan assay.Wethenanalysedtheassociationbetweenthepresenceofthegeneticvariantanddiseaseseverity and viralload.WedidnotobserveanycorrelationbetweenthepresenceofTMPRSS2geneticvariantandthe severity ofthedisease.However,weidentifiedasignificantassociationbetweenthep.Val160Metpolymorphism and theSARS-CoV-2viralload,asestimatedbytheCtvalueofthediagnosticnucleicacidamplificationtest. Furthermore, weobservedatrendofassociationbetweenthepresenceoftheCalleleandthemortalityratein patients withsevereCOVID-19. Conclusion: Our dataindicateapossibleassociationbetweenTMPRSS2p.Val160MetpolymorphismandSARS-CoV- 2 infectivityandtheoutcomeofCOVID-19.
