Varian Genotip Gen NRAMP1 dan Ekspresi Protein NRAMP1 Pada Perawat dan Penderita Tuberkulosis Paru.

Abstract
The relationship between natural resistance associated macrophage protein-1 (NRAMP1) genotype variant and susceptibility or resistance against pulmonary tuberculosis has not yet been explored in Indonesia. In order to prove this relationship, a research was conducted on 112 total samples consisting of 69 pulmonary tuberculosis patients and 43 pulmonary ward nurses from May 2006 until April 2007, as a observational - crossectional analytic design.
This study was focused on observing the NRAMP1 gene of genotype variant of D543N, 3”UTR, INT4 using PCR-RFLP method and the NRAMP1 protein expression in macrophage using immunocytochemistry in both tuberculosis patients and pulmonary ward nurses group. The data obtained were analyzed towards difference, interaction and relationship between the NRAMP1 gene of genotype variant of D543N, 3’UTR, INT4 and the NRAMP1 protein expression, in both groups.
The result of Chi-Square analysis the NRAMP1 gene of 3 UTR genotype of TGTG+/+, TGTG+/del, TGTGdel/del variant between in nurses with in pulmonary TB patients revealed a significant difference with p=0,007 (p<0,05). The result of Oneway Anova analysis between the NRAMP1 protein expression with the NRAMP1 gene of 3 UTR genotype of TGTG+/+, TGTG+/del, TGTGdel/del variant in pulmonary TB patients revealed a significant difference with p=0.000 (p<0.05).
In conclusion, the NRAMP1 gene of 3’UTR genotype of TGTGdel/del variant occurs more frequently in tuberculosis patients than in nurses, and this variant had degrade of the NRAMP1 protein expression
