Abstract

The major object of this research was : The Production of Polyclonal ArHibody from rabbit that was immunized with local spec if ic prot ein S.m 1 (c). The production of pol yclonal antibody was tested against lo calS. m 1 (c) colonization (in vitro) . ,__ Methods, the cell wall lo cal S. m 1 (c) was fractinated by sonication to extract the protein . The protein molecular weight was determined by electrophoresis (SDS-PAG E) . The specific protein was further checked by Western blotting. The first antibody used was rabbit antiserum from rabbit that was immunized with lo cal S. m 1 (c). The second antibody used was fragment lgG goat anti rabbit conjugate . The immunoblotting resulted to get the most characteristic protein of /veal S. m 1 (c) ; 67 k Da molecule in weight. This protein was further processed by purification to produce a vaccine to be injected in the rabbit (800 ~g / ml per injection) After three boosters injection , serum was collected from rabbit that reached a peak of titer 1/240 .000 . Polyclonal antibodies against the antigenic protein of lo calS. ml(c) was tested in vitro using the modified cy linder and we ll methode (Wistreich & Leichtman, 1980)
