Abstract
Water obtained from handpiece dental unit is densely populated with microoorganisms. The type of microorganism isolated includes both envorimental microrganisms and opportunistic pathogen microorganisms, thus make this water a potential source of infection. Flushing dental unit waterlines for several minutes reduces viable bacterial counts. The aim of this study is to identify various kind of microorganisms in dental unit waterlines which is supplied by municipal water (PDAM) and distilled water; to investigate viable bacterial counts in both sources; and to investigate the effect of 1, 2, 3, 4, 5 minutes flushing in reducing viable bacterial counts. The microorganisms was identified by Gram staining and Schaeffer Fulton staining under microscope observation, inoculation in specific media and
biochemical tests. E. coli was identified by Multiple Tube Method 1-5-5 modification. Viable bacteria in both water sources and after 1, 2, 3, 4, 5 minutes flushing wer'-! counted by droplett method in BHI agar. The result showed that many kinds of bacteria could be isolated aerobic and anaerobically from dental unit water sources, including Pseudomonas aeruginosa dan Escherichia col!: Analysis of Variance showed significant difference in viable bacterial counts between municipal water (PDAM) and distilled water (p<0,05). Municipal water contains viable bacterial counts higher than distilled water. Flushing dental unit waterlines for 1, 2, 3, 4, 5 minutes in both water sources reduces viable bacterial counts significantly (p<0,05). Five minutes flushing reduces viable bacterial counts maximally.
