Abstract

An investigation was undertaken to determine the effects of elevated temperatures on testicular cells and to seek distribution of stress proteins (HSP 70) associated with exposure of testes to heat stress. In this experiment, scrotal regions of rats were exposed to 40.0 degrees C H20 bath for 60 minutes.Rats were killed on various days (days 1, 4, 7, 14, 21) after exposure. Testicular cells changes were determined by Hematoxilin-Eosin staining and distribution of HSP 70 were sought by immunohistochemistry. Testicular weights of rats decreased significantly day 4 through 21 (p < 0.05 ). Also significant decrease in diameter and epithel thickness of seminiferous tubules were seen on days 1 — 21 ( p < 0.05 ).Vacuolization of the germinal epithelium was noted synchronously with the onset of germ - cell loss. HSP 70 were visible in both control and experiment group. When compared to control, distribution of HSP 70 on days 4 through 21 were more extensive. These results show that rats spermatogenesis is disrupted by scrotal exposure to enviromental temperatures several degrees over normal physiological temperature and HSP 70 expression in germ cells plays a specific role in spermatogenesis as well as in stress response. Further investigation of the biochemical mechanisms involved in testicular temperature may provide strategies to adress a vatriety of male reproductive issues.
