Isolasi dan Karakterisasi Protein Imunogenik Larva Stadium II Toxora Cati Isolat Lokal.

Abstract
The aim of this study was to get the purified protein with the high specificity. immunogenicity and antigenicity. It can be used to produce the monoclonal antibody and also as a kit of toxocariasis diagnostic. This study was divided in some steps as follows: isolation of second larvae and adult worm; production of whole extract and polyclonal antibody; analysis of protein antigen; characterization of protein antigen by Western blot; and purification of protein antigen by preparation of gel electrophoresis. Structural characterization of the second larvae of Toxocara cati by sodium dodecyl sulphate polyacrilamide gel electrophoresis (SDS-PAGE), showed that the crude extract consisted of seven polypeptides of 133.7 kDa, 96.6 kDa, 86.1 kDa, 60.7 kDa, 40.0 kDa, 30.3 kDa and -24.0 kDa. By immunoblotting, the cross-reaction antigens of F. gigantica were recognized by anti-second larvae of T. cati antisera was 133.7 kDa. Anti-second larvae of T. call antisera was detected by 7'. cati and T. vitulorum larvae and both adult stage was 30.3 kDa. Eventhough, the anti-second larvae of T. cati antisera was detected by only of both T. cati and T. vitulorum second larvae, was 24.0 kDa.
