Abstract
chitosan with carbonate apatite (CA). The purpose of this study was to fabricate this composite and to evaluate carbonate apatite-chitosan scaffolds (CA-ChSs) from a standpoint of cell proliferation using MC3T3-E1. Chitosan scaffolds (ChSs) which contained of 25, 50, 75, 100, 125, 150, 200 and 400 mg chitosan were fabricated by the following procedure (100D, YSK, Japan) was dissolved into 5 ml of 2% acetic acid, shaked for 15 min, then neutralized with 15 ml of 0.1 M NaOH solution. After centrifugation at 1500 rpm for 10 min, excess water was removed and then chitosan gel was packed into the molds (diameter: 5 mm, height: 2 mm). The molds were frozen at -80°C for 2h and dried in a freeze dry machine for 24h for sponge body. The sponges were subjected to UV radiation for 2h. To fabricate CA-ChSs, 200 mg chitosan was selected. After neutralization, 10, 50, 100, 200 and 300 mg of 0.06 M CA were added into the 200 mg ChSs. The structure of CA-ChSs was observed by scanning electron microscope (SEM). Mouse osteoblast-like cell (MC3T3-E1) proliferation in this scaffold was investigated for 1, 7 and 14 days. The result showed that three dimensional porous structures of CA-ChSs with CA powder attachment were clearly observed by SEM. Proliferated cell numbers in CA-ChSs were significantly higher than those in ChSs (control) in each stage for 1, 7 and 14 days (p< 0.05). In conclusion, it may suggest that CA-ChSs is a possible candidate for bone tissue engineering.
