Abstract

The purpose of the research is to see the effectiveness from washing whole membrane to level of outer membrane protein Brucella abortus S-19 is measured by Bradford Protein Assay method. The sample being used in this research is the stock of whole membrane active bacteria Brucella abortus S-19 from Pusvetma Surabaya. To ensure its purity, bacteria was grown at Potato Agar. The sample was washed using PBS followed by centrifugation for 25 minutes with 5000 rpm speed to get the residue bacteria. Sonication of the whole membrane is done with an ultra homogenizer, which aimed to break the wall of a bacteria cell in order to get the outer membrane protein (OMP). Then the OMP is added to PBS and it was centrifugated. The level of the OMP was counted by adding the standard formula BSA and reagent Bradford to be measured by Bradford protein assay with 595 nm. The results of research showed that the numerical of Bradford counted were 3,84 g"/o and 3,42 g"/o at once washing, 3,45 g"/o and 3,39 g"lo at twice washing and the last at three times washing showed that the results 3,25 g"/o and 3,24 g"lo. Result showed that the number of washing did not give effect to level of the protein from OMP Brucella abortus S-19.
