Abstract
The objectives of this study is to determine the growth of rabies virus street strain from the different geographical area of Indonesia (the island of Sulawesi, Nusa Tenggara, Kalimantan and Sumatera), the molecular weight of N and G proteins and the immunogenecity of the N and G proteins, was compared between street strains and virus from laboratory (fixed and CVS strain). The second study is to determine the nucleotide sequence variation of the fragment N and G genes for the correlation of homology group. This study was divided on several stages, including biological characterization of rabies virus in neuroblastoma cell lines, identification of virus by indirect sandwich ELISA and direct FAT, and protein characterization by SDSPAGE and immunoblot. The immunogenecity of protein coding was applied in mice. Molecular characterization by RT-PCR assays and genetic analysis was performed in PCR products. The nucleotide sequences in this report have been submitted to GenBank by GenMac version 8.0. The results showed that the growth of rabies virus strain from Sulawesi, Nusa Tenggara, Kalimantan, Sumatera and laboratory strain were significant different (p<0.01) of optical density antigen at the first and third passage. The molecular weight of the protein N and G protein was 56.2-58.2 kDa and 66.7-69 kDa in respectively. Immunogenecity test of proteins in mice showed significant different (p<0.01) of optical density antibody induced by N and G protein. The PCR product analysis of 406 bp fragment of the N gene and 210 bp fragment of the G gene represented the molecular study done in Indonesia. Comparison of the sequence of rabies virus street isolate with the sequence of reference virus for GenBank showed 79.7-96.8 % (N genes) and 87.5-93.6% (G genes) homology.
