Abstract

The growth of prostatic tissue is stimulated by testosterone. The incidence of benign prostatic hyperplasia (BPH) is higher in old age when the testosterone level is low. At this time luteinizing hormone (LH) is higher than normal and estrogen concentration is relatively higher than testosterone. This discrepancy stimulates our curiosity to investigate the pathobiology of BPH. Prostate specific antigen (PSA) as an imunogen is a product of prostatic gland. The effects of estrogen and LH on the prostatic components (androgen receptor, gland, smooth muscle, interstitial fibers), PSA and immune response were studied. We used young male monkeys (Macaca fascicularis) for this experiment. The method used was pure experimental with the pretest-posttest control group design. Samples were divided into 3 groups. 12 monkeys for esch group. Estrogen was given to the first group, LH to the second group and the third one was the control group. The treatments were given for 4 months. The variables detected were hormones concentration in plasma, immunologic components in blood, PSA and the prostatic tissue components. In this experiment we found that androgen receptors were increased in the prostatic tissue after estrogen and LH treatment. The abundant androgen-receptor complexes caused more growth stimulatory factor production and the proliferation of prostatic components were stimulated. The prostatic weight and volume increased. In estrogen group, PSA concentration as an inununogen raised and caused a worse inunune respons. In LH group, PSA concentration also raised, but much lower than in estrogen group. This low concentration of PSA caused a better immune response than the inunune response of the estrogen group. It was concluded that estrogen and LH played important role in the enlargement of prostatic gland and in altering the immune response. The result of this experiment was hoped to be used for the rational prevention and treatment of BPH and for further experiments in discovering the pathobiology and inununopathology of BPH.
