Abstract
This research was aim to know the influence of osteopontin to increase the quality of bull spermatozoa in order to improve in vitro fertilization rate. Sample used in this research were ovarium of Friesian Holstein cows sloughtered in Bajangan sloughterhouse, Gondangwetan Pasuruan. The mediun used to mature oocytes was HTF added with 0,01 μg/ml, FSH 0,01 μg/ml LH, 3% bull with two groups treatment (PO without osteopontin supplementation 0 μg/50 million spermatozoa) and (Pl with osteopontin supplementation IOμg/50 million spermatozoa). Before In Vitro fertilization, post swim up protein identification of frozen semen without and with supplementation (PO and Pl) was conducted protein molecular weight was evaluated by SDS-PAGE electrophoresis method. The differenses between protein profile evaluation of PO & Pl was caused by the effect of OPN protein supplementation in Friessian Holstein (FH) bulls frozen semen (Pl). In the Pl groups 13 protein bands were found while PO groups only 11 protein bands were found. The result of protein identification on post swim up frozen semen ofFH bulls in PO and Pl groups showed that OPN molecular weight was 61,66 kDa. Fertilization rate evaluated by release of second polar body or formation of 2 pronucleus after fertilization process. There was significant difference between group without and with OPN supplementation (P < 0,05). OPN suplementation in frozen semen has positive effect to fertilization rate of cow oocyte in vitro.
