Abstract

This experimental study has been carried out to investigate antifertility effect of flavonoids of Justicia gendarussa Bunn.f leaf on penetration inhibition of mouse spenn in vitro fertilization (IVF). The aim of this study was to detennine major flavonoid compound structure of n-butanol fraction that influences the penetration inhibition by decreasing intracellular spenn hyaluronidase. Five steps of investigation were carried out: I). Extraction and fractination with n-hexane, methanol, chlorofonn and n-butanol respectively; 2). in vitro and in vivo activity test of penetration inhibitation by IVF method; 3). isolation, separation, purification and structure elucidation of the flavonoid compounds; 4). In vitro activity test of major compound isolate by lVF methods; 5). in vivo spenn hyaluronidase activity test. The results of this study showed that in vitro activity was 75 ppm and in vivo activity (per oral) was 15 and 30 mw20 g BW. /days of n-butanol fraction on penetration inhibition by IVF methods, Hyaluronidase mouse epididymal spenn activity was decreased whith persistent synthesis of hyaluronidase. n-Butanol fraction consists of the flavonoid compounds with the same molecule weight by LC-MS. and only two were isolated. In vitro, effect of major compound was active in 100 ppm concentration by IVF methods.
