Abstract

"To guarantee the Indonesian folk medicine’s safety and efficacy, it must be made as phytopharmaceutical dosage form, which one of its step is quantify active compound by valid method. One of Indonesian folk medicine is guava fruits (Psidium Guajava L.). Quercetin was used as a marker because of its activity, such as antivirus, immunomodulator and antihistamine. TLCDensitometry method was developed to evaluate and validate the assay of quercetin. TLC of quercetin on silica gel F 254 with chloroform-glacial acetic acid-aquadest (7:3,3:0,8) as mobile phase got resolution 2,8. Qualitative identification was done by comparing sample’s Rf and spectrum profile with quercetin reference standard. Quantification by densitometry at λ 375 nm. The limit of detection (LOD) of quercetin was 0,47 ng. The limit of quantitation (LOQ) of quercetin was 1,41 ng. The correlation coefficient of linear calibration for quercetin was 0,996. The coefficients variation for ruby guava fruit, sweet guava fruit, and Bangkok guava fruit were 2,45%, 2,13% and 2,56%, respectively. The mean concentrations of quercetin for ruby guava fruit, sweet guava fruit, and Bangkok guava fruit were (16,67 ± 0,408) ppm, (19,72 ± 0,420) ppm and (3,17 ± 0,081) ppm, repectively. The recovery of quercetin was 90-110%."
