Abstract

Shiga toxin is a toxin produced by STEC bacteria, one of which is the bacterium Escherichia coli 0157: H7. Shiga toxin is a toxin that can cause Haemorrhagic Colitis (HC) and Hemolytic Uremic Syndrome (HUS) in humans. In this study explains the the sourcing and characteristics of shiga toxin genes isolates Esherichia coli 0157: H7 on fresh milk taken at from milk can at the farm located in the Surabaya city. 40 samples of milk in the milk can there were 24 positive samples contained Esherichia coli that had been planted in the EMBA media and was confirmed by lndol test. 24 samples are then conducted confirmatory test by multiplex-PCR using a primer for the specific H7 and shiga toxin 2 gene, and then Multiplex-PCR results were obtained 3 samples showed the presence of the band stx2. Three of the test sample to look for nucleotide by sequencing method. After sequencing, we flooking for the homology of the sample using Bioedit software called ClustalW2, and obtained the results that have a kinship score of 91.86 Esherichia coli 0157: H7 and 76.45 stx2. The conclusion of this study was that the milk taken from dairy farms in Surabaya city be found the bacteria Escherichia coli 0157: H7 but does not cause HC and HUS in humans because stx2 produced is different.
