Abstract
Hesperetin is an active drug ingredient that belongs to the class II BCS drug class, which means it has low solubility and high permeability. This condition makes hesperetin has a low dissolution rate and bioavailability. Therefore, the dissolution rate needs to be increased to optimize the dosage used. One of the methods that can increase the dissolution rate is the nanosuspension drug delivery system that uses the principle of particle size reduction to nanometer, which will be dispersed in water and stabilized with Kollicoat® IR polymer. The method used for particle size reduction is the Top-Down Technology method which relies on reducing the size of larger particles to achieve nanometer sizes, such as milling techniques. Currently, nanosuspensions are widely used in solid dosage forms because of their acceptability, ease of distribution, and practicality of use. This research is a preliminary study in the manufacture of solid pellet preparations coated with hesperetin nanosuspension with the fluid bed coating method. So as a preliminary study, this research will examine the effect of giving different concentrations of Kollicoat® IR to the dry form of hesperetin nanosuspension. The selection of the Kollicoat® IR polymer was based on its advantages of having high water solubility, its good viscoelastic properties as a suitable pellet coating excipient, and its nature as an amphiphilic polymer. The dissolution test was carried out with the kontrol variable Nanosuspension, while the independent variables were dried nanosuspension and redispersed dried nanosuspension. In other words, this research has two kinds of nanosuspension, the first one is the dried one, and the other one is dried then redispersed one. This treatment was aimed to determine whether there was a significant difference in the addition of Kollicoat® IR concentration in dry hesperetin nanosuspension
