Abstract

The purpose of this research is to determine the total number of bacteria in raw milk at BUMDes Sinar Mulya Tulungagung working area by Total Plate Count (TPC) method compared to SNI 3141.1:2011. The total number of bacteria was tested by the TPC method at the Regional Health Laboratory, Tulungagung Health Office. The sampling technique used purposive sampling method. This experiment used 30 samples of raw milk collected from 30 farmer members of BUMDes Sinar Mulya Tulungagung. The solution used as diluent was Sodium Chloride 0,9%. This research used the Pour Plate method procedure that was inoculated 1 ml of milk sample used a sterile pipette into a labeled sterile petri dish. As much 20 ml of Nutrient Agar medium (cooled to 45 – 50 °C) was poured, then homogenized by rotating the petri dish horizontally to make the media spread and let it solidify. Incubated the petri dish into incubator with upside position for 24 hours at 37 °C. Microbiological analysis results report was reported used a standard called the Standard Plate Count (SPC). Total Plate Count result showed that the total numbers of bacteria in raw milk at BUMDes Sinar Mulya Tulungagung working area was 4,3 x 105 CFU/ml that met the SNI 3141.1:2011.
