Abstract
In Indonesia, Ruta angustifolia L. is known as a traditional medicine to cure as jaundice (liver disease) (Wahyuni et al., 2014). Isolates from the ethanol extracts of R. angustifolia L. chalepin and pseudane IX were reported to have anti-hepatitis C activity, containing IC50 with total of 1.7 ± 0.5 μg/mL and 1.4 ± 0.2 μg/mL and no cytotoxicity (Wahyuni et al., 2014). Moreover, the combination test with simeprevir (SMV) and telaprevir (TVR) was found to have a synergistic effect (Wahyuni et al., 2020). The 70% ethanol extract of Ruta angustifolia L. incorporates rutin (0.06% ± 0.34) which has anti-HCV activity and can be developed into anti-HCV drugs (Wahyuni et al., 2021). R. angustifolia L. is possibly be developed into a phytopharmaceutical drug, however environmental variables cannot guarantee compound content of plantation products to be constant. Meanwhile, phytopharmaceutical products require standardization to ensure safety, quality, and efficacy. An essential parameter to undertake the quality of a standardized herbal medicinal product or phytopharmaceutical is through standardization (Depkes, 2000). The study probed specific and non-specific standardization parameters on 70% ethanol extract of the herb R. angustifolia L. The results of determination of specific standardization parameters included the ascertainment of ethanol solubility extract content and the water solubility extract of 15.26% and 80.19%. The chromatography analysis by thin layer chromatography was demonstrated to possess flavonoid compounds and have the some Rf value with the standard compound of rutin. Further analysis by HPLC have not given optimal separation. Non-specific parameters were obtained total ash content (14.41% ± 0.2) and water content (9.244% ± 0.2).
