Abstract
The potential of milkfish in East Java is very high, in 2019 the production of milkfish in East Java Province was recorded at 171,360.75 tons/year (Statistics-KKP). High fish production requires good handling technology because fish are prone to decay. Lack of attention to food security such as unhygienic handling can cause health problems. The most worrying is microbiological contamination. One of them is E. coli serotipe O157 H7 who cause of foodborne illness in human. The objective of this study was to identify E. coli serotipe O157:H7 in milkfish at 4 fish market in east java (Gresik, Sidoarjo, Lamongan and Surabaya) in 20 sample tested by Total Plate Count (TPC). Only sample with exceeded of maksimum contaminant will be tested by PCR. For first identification, the sample will be tested by TPC, The result showed 7 of the sample have exceeded the maximum contaminant according to Standart Nasional Indonesia (SNI 7388:2009). Subsequently, the positif samples were tested by EMBA (selective and differential medium). The results showed that 5 of 7 samples are contaminant by E coli. the positif samples with E. coli were tested with PCR to knowing the serotipe O157:H7 with primer product target gen 239 bp. The result are 1 of 5 samples is positif patogen serotipe O157:H7, the positif sample is Surabaya water sanitation.
