Abstract

Introduction. Alveolar bone defects which caused by periodontal disease, another pathologic disease requiring regenerative therapy using bone grafts. Anadara granosa shell graft (AG) has osteoconductivity so it can be a scaffold to attract pluripotent cells that support bone healing process. Purpose. To investigate the new graft material using Anadara granosa shell and mechanism to accelerate and increase osteogenesis in mandible. Methods : Total of 30 male rats Rattus norwegicus aged 6 months with body weight 250-300 grams were randomly devided into 6 groups with 5 rats each group. All samples got it’s mandibular right incisors extracted, curettage and irrigation. The group consist of : treatment group with AG, treatment group with xenograft, control group untreated graft, after one week rats were sacrificed for osteoblast, osteoclast, FGF-2, VEGF, OPG analized. Treatment group with AG, treatment group with xenograft, control group untreated graft, after two weeks rats were sacrificed for osteoblast, osteoclast, BMP-2, OCN analized. Result : AG shell graft containing 44% calcium carbonate, 33% calcium hydroxide and 23% calcium phosphate could increase the expression of FGF-2, VEGF, OPG, osteoclast and reducted osteoblast at 7th days. It could increase the expression of BMP-2, OCN, osteoblast cells and reducted of osteoclast at 14th days. Conclusion : AG shell graft has potential as a graft material in the mandible osteogenesis process with increased osteoblasts, decreased osteoclasts and increased OCN via BMP-2.
