Abstract

Ethnopharmacological relevance: Justicia gendarussa Burm.f. has been traditionally used as male contraception in Indonesia. Aim of the study: Acute and subchronic oral toxicity studies of Justicia gendarussa Burm.f. in rodents were performed in the present study in order to evaluate its safety. Materials and methods: In acute toxicity study, 70% ethanol extract of gandarusa leaves was administered by oral gavage to Balb/c mice (5 control and 5 treatment) at a dose of 2000 mg/kg. In subchronic toxicity study, the extract at doses of 40, 200, and 1000 mg/kg/day were given orally to groups of male rats (10 rats/dose) for 90 consecutive days. Results: The extract at a dose of 2000 mg/kg produced no treatment-related signs of toxicity or mortality in any of the animals tested during 14 days of the study. In the repeated dose 90-day oral toxicity study, there was no significant difference in body weight between the control and all treatment groups. At dose 40 mg /kg BW, ethanol extract 70% J. gendarussa leaves did not affect SGOT, SGPT, and Creatinin, but affect BUN. At a dose of 40 mg / kg BW of ethanol extract 70% of J. gendarussa leaves did not affect all histopathological parameters of the liver and kidneys. Conclusions: These results suggest that further research is needed to ensure its safety for clinical study.PCA, SIMCA
