Abstract

Hesperetin is an anticancer drug that has low solubility in water. The micelle delivery system is efficient for the passive targeting of tumor cells and increases the solubility of poorly water-soluble drugs. In this study, hesperetin combination micelles were prepared in TPGS and Poloxamer P84. Micelles cannot be stored for a long time in a liquid state. The high moisture content causes the micelles to damaged easily. Therefore, solidification was carried out using the freeze-drying method. This study aims to evaluate the effect of maltodextrin as a protectant on the physical characteristics of the hesperetin combination micelles in the freeze-drying process. The four formulas used in this study were without maltodextrin (F1), 10% maltodextrin (F2), 15% maltodextrin (F3), and 20% maltodextrin (F4). The results of the particle size evaluation showed that the particle size was not affected by the increase in maltodextrin concentration. The particle size after freeze-drying was the same as before freeze-drying. The moisture content in F4 was found to be 9.5%. The results of the re-disperse test showed that F4 had the best redispersion time. The results of the drug entrapment and drug loading showed that drug entrapment and drug loading were greater as the concentration of maltodextrin increased. However, there was a decrease after the freeze-drying process. Based on these results, F4 is the best formula because it has the best freeze-drying characteristics, the smallest particle size, the best dispersibility, and the highest drug entrapment and loading.
