Abstract

Introduction: Pterygium is a degenerative illness defined by the formation of fibrovascular tissue on the ocular surface. The recurrence of this condition, which is accompanied by inflammation and increasing fibrovascular growth, is regarded as its key therapeutic challenge. Epigallocatechin gallate (EGCG) has demonstrated anti-inflammatory, antioxidative, and antiangiogenic benefits in many models of ocular illness. Our study aimed to determine and compare the efficacy of EGCG with MMC on pterygium fibroblast proliferation and expression of TGF-B. Methods: Pterygium fibroblast from tissue explants during pterygium surgery was cultured and treated with EGCG 100 µM, 150 µM and MMC 0.4 mg/mL. Proliferation was assessed using the CCK-8 assay. The expression of TGF-? was determined with immunofluorescence staining and the intensity measurement was done using ImageJ software. The results between groups were analyzed using ANOVA test and post hoc test. Results: EGCG showed a concentration-dependent inhibition of TGF-B expression and fibroblast proliferation. EGCG group showed significantly inhibited expression of TGF- B compared to the control group (p<0,001), where there 150 µM EGCG group showed lower TGF-B level than 100 µM EGCG group by 52826 ± 6326,313 and 72958,444 ± 6399,822 , respectively, at 48 hours. MMC 0,4 mg/mL also showed significantly reduced expression TGF-? compared with control group (p<0,001). EGCG and MMC also significantly inhibited pterygium fibroblast proliferation (p<0,001) Conclusion: EGCG and MMC inhibits pterygium fibroblast proliferation and expression of TGF-B. These findings showed that EGCG represents a potential therapeutic agent to prevent the recurrence of pterygium after surgery.
