Abstract

Sendang Biru merupakan salah satu kawasan pesisir yang menjadi sentra industri perikanan tangkap di Kabupaten Malang Jawa Timur. Hasil tangkapan ikan yang banyak diperoleh dari pantai ini salah satunya adalah ikan kakap. Ikan kakap merupakan salah satu jenis ikan demersal dari famili Lutjanidae. Famili Lutjanidae yang tersebar di seluruh dunia saat ini memiliki 123 spesies dalam 21 genus, salah satunya yaitu genus Lutjanus. Hingga saat ini, pencatatan data produksi perikanan tangkap untuk ikan kakap di Malang masih sangat terbatas pada jenis tertentu. Identifikasi morfologi masih sulit mendapatkan hasil yang akurat karena banyaknya kemiripan antar spesies yang diamati. Oleh karena itu, perlu dilakukan identifikasi secara molekuler dengan DNA barcoding untuk mendapatkan hasil identifikasi ikan kakap yang lebih akurat hingga tingkat spesies. Penelitian ini bertujuan untuk mengetahui jenis-jenis ikan kakap yang didaratkan di Pelabuhan Pondokdadap Sendang Biru, Malang Selatan menggunakan pendekatan molekuler pada marka gen Cytochrome Oxidase subunit I (COI) dan merekonstruksi pohon filogenetik ikan kakap berdasarkan data sekuens DNA serta mengetahui status konservasinya. Penelitian ini merupakan penelitian observasi dengan memperoleh data melalui pengamatan morfologi ikan kakap serta pengambilan sampel daging ikan untuk identifikasi molekuler. Hasil identifikasi ikan kakap berdasarkan rekonstruksi pohon filogenetik dapat diketahui bahwa sampel L. bengalensis memiliki kedekatan dengan L. rufolineatus sedangkan L. gibbus, dan L. erythropterus masing-masing membentuk clade terpisah dari dua jenis Lutjanus sebelumnya. Berdasarkan status konservasinya pada IUCN, keempat spesies ikan kakap yang ditemukan masuk kedalam kategori Least Concern (LC) sedangkan berdasarkan status perdagangan pada CITES, keempat spesies ini masuk dalam kategori Not Evaluated (NE) atau tidak dievaluasi. Sendang Biru is one of the coastal areas that are the center of the capture fisheries industry in Malang Regency, East Java. One of the catches of fish obtained from this beach is snapper. Snapper is a type of demersal fish from the Lutjanidae family. The Lutjanidae family which is spread throughout the world currently has 123 species in 21 genera, one of which is the Lutjanus genus. Until now, the recording of capture fisheries production data for snapper in Malang is still very limited to certain types. Morphological identification is still difficult to obtain accurate results because of the many similarities between the observed species. Therefore, molecular identification is necessary to determine the types of snapper in this area and their conservation status. This study aims to determine the types of snapper landed at Pondokdadap Fishing Port Sendang Biru, South Malang to the species level using a molecular approach to the Cytochrome Oxidase subunit I (COI) gene marker and reconstruct the phylogenetic tree of snapper based on DNA sequence data and determine its conservation status. This research is an observational study by obtaining data through observing the morphology of snapper and taking fish meat samples for molecular identification. The results of the identification of snapper based on reconstruction phylogenetic tree showed that the L. bengalensis samples were closely related to L. rufolineatus, while L. gibbus, and L. erythropterus each formed a separate clade from the previous two Lutjanus types. Based on their conservation status at IUCN, the four snapper species were found are in the Least Concern (LC) category, while based on their trading status at CITES, this four species are in the Not Evaluated (NE) category.
