Abstract

Andrographis herbs (Andrographis paniculata Nees.) and Mahogany seeds (Swietenia mahagoni Jacq.) have known as antidiabetic. These studies were carried out to evaluate acute and subchronic toxicity toward the mixture of 70% ethanol extract of Andrographis herbs and Mahogany seeds granule. These studies aimed to fulfill qualification of phytopharmaceutical products registration. The experimental research design were used 25 male balb/c mice for acute toxicity with 20 male wistar rats and 20 female wistar rats for subchronic toxicity. The acute toxicity were divided into five treatment groups randomly with a dose of 5000, 2000, 300, 50, and 5 mg/kg BW via a stomach sonde one time only for 24 hours and observed their death and toxic symptomps along 14 days. The subchronic toxicity were divided into four groups randomly: one group (K) were given CMC Na suspension, and three treatment groups with a dose of 67.5, 135, and 270 mg/kg BW via a stomach sonde times per day for 28 days and observed their clinical parameters such as hematology, biochemistry clinic, and relative vital organ weight and also toxic symptomps. The acute toxicity data were analyzed by probit analysis and subchronic toxicity data were analyzed with One-Way Anova Test followed by Post-Hoc Test. The acute toxicity result showed there were no death mice and toxic symptoms. It can be determined that the LD50 of 70% ethanol extract granule combination was more than 5000 mg/kg BW, which are relatively harmless drug categories. Then the subchronic toxicity result showed these combination didn’t cause toxic effect on male and female rats.
