Abstract
Comparative study between conventional and vitrification methods on man
with normozoospermic sperm towards morphology, motility, viability and
chromatin condensation of spermatozoa". (under guidance of Prof. DR. H.
Soehartojo Hardjopranjoto, Drh., MSc. and dr. Aucky Hinting Ph.D.).
Vitrification method is an alternative for cryopreservation of spermatozoa
which is continuously studied and developed because the technique is more
efficient, cheaper, simpler and easily carried out, compared to the
conventional method. The purpose of this study is to observe the results of
the vitrification as well as to examine the quality of spermatozoa after
vitrification, so that it can be used as a model for cryopreservation of sperm
especially for the ART (Assisted Reproductive Techniques).
The study applies cryoprotectants glycerol 15% + HEPES + Human Serum
Albumin (Medium Freeze, Fertipro, Belgi). The sample is normozoospermic
sperm - 10 ejaculates treated in three ways. The first is observed before
cryopreservation, and the other two groups are put in mini straw 0,25 mi.,
then preserved in frozen condition using conventional and vitrification
methods. After being frozen for 24 hours, the sperm are thawed, then stained
and observed under microscope. The parameter is the percentage of normal
morphology, motility (a+b), viability and chromatin condensation of
spermatozoa.
