Abstract

This research was aimed to investigate the colour changing of Escherichia coli colonies in artificial medium that was made from beef extract, mung beans juice, lactose and phenol red. The media were made by mixing 1.6 % agar base, 0.8 % beef extract, 20 % mung beans extract, 0.2 % lactose, phenol red solution and aquadest ad 1000 ml. The isolate of Escherichia coli were inoculated onto the media with five repetitions. As control, Escherichia coli were inoculated onto the media that were made from beef extract and mung beans juice, without lactose and phenol red. The whole media were incubated at 37 oC for 24 hours. The result showed that the Escherichia coli colonies appeared brownish-white and shiny. The growth of Escherichia coli on the treatment medium gave colour change on the medium from red to yellow. The colonies on the treatment medium appeared yellow and shiny. Based on the result, the artificial medium that was made from beef extract, mung beans juice, lactose and phenol red showed that the medium quite good for Escherichia coli and showed a colour changing by Escherichia coli that ferment lactose.
