Abstract
H-Y antisera have a high activity of anti-male so that it can eliminate Y-chromosome spermatozoa that have faster motility because of DNA content and size of the head is lighter than X-chromosome spermatozoa. H-Y antisera mechanism of action is through immunologic reaction in which the H-Y antisera as antibody (Ab) will bind H-Y antigen (Ag) contained in the spermatozoa. Immunological reaction in the form of bonds H-Y Ag and H-Y Ab was then that remains is pure X-chromosome spermatozoa. The purpose of this study was to determine the effect of the addition of H-Y antisera against spermatozoa motility and viability post-thawing of Merino sheep. In this study using 5 treatments with 4 kinds of H-Y antisera dilution: P1 (H-Y antisera 2X104 dilution), P2 (H-Y antisera 4X104 dilution), P3 (H-Y antisera 6X104 dilution), P4 (H-Y antisera 8X104 dilution) and P0 (0.25 cc post-thawing Merino sheep’s semen) as a control with the time of incubation for 30, 45, and 60 minutes. Sperm motility was observed with sperm vision, sperm viability was observed from eosin nigrosin stain with microscope. Quantitative data were analyzed using ANOVA and continued by Tukey test on 5% significance level. The result showed that the 4 kinds of H-Y antisera dilution affect sperm motility but had no effect on sperm viability. Based on time incubation H-Y antisera affect sperm viability but had no effect on sperm motility.
