Abstract

Problems usually occurs on frozen sperm are the effects cold shock towards the cells and intracelluler changes as the result of the ice crystal forming. Theese ice crystal can cause accumulation of electrolite and other soluble substances inside the cells that could damage intracellular part of sperm. This intracellular damage is hard to detect but can cause death of the sperm. Until now, this case has rarely been revealed on molecular biology. The purpose of this research was to compare profile of band dna dairy cattle FH sperm fresh and frozen. Each sample were checked microscopically and macroscopically before being isolated. Profil of band DNA and total base pair can be observed with elektroforesis agarose gel. The dna band profile appeared in paralel line. Based on the calculation, both of the fresh n frozen sperm had the same anmount of total dna which was 10238 bp. There was no significant changes on the sample. So that it could be assumed that the freezing procees and cold shock did not affect the amount of total dna on FH bovine sperm.
