Abstract
Growth differentiation factor-9 (GDF-9) is a 51 kDA glycoprotein as a part of Transforming growth factor-beta (TGF-β) subfamily secreted by oocytes. GDF-9 plays role in oocyte maturation by stimulating granulosa cells proliferation and inhibiting differentiation. The purpose of this study was to identify GDF-9 expressed in bovine oocytes that were collected from follicle with 2-5 mm diameter. This study used a method based on specific immunoaffinity binding between avidin molecules biotinilated into the secondary antibody (avidin-biotin complex). GDF-9 was expressed in 38 oocytes out of 48 collected oocytes (79.17%). Expression was characterized by dark-brown colour due to chromogen visualization of DAB in the SA-HRP complex boundaries. GDF-9 down-regulated oocytes were methylen green counterstained
