Abstract
Sex Identification in parakeet (Melopsittacus undulatus) is very important to help conservation and breeding efforts in order to preserve and increased the population of parakeets. Especially in monomorphic bird, that was difficult to distinguish between male and female. Molecular-based technology approach with PCR (Polymerase Chain Reaction) method can be applied to amplify intron length that can be distinguish between CHD-W and CHD-Z and allowed to distinguish gender by using P2 and P8 primers, this method can be used to determine the sex of the parakeet appropriately and accurately using feather samples. This research was conducted in two ages five months parakeets, using three to four feathers that have been extracted and amplified by PCR (Polymerase Chain Reaction), and read using 1% agarose gel electrophoresis, the male appeared one band, and female appeared two bands. gender verification has been done by performing surgery on each bird to saw the morphology of the reproductive organs, the result of reproductive organ morphology and electrophoresis showed that the parakeet was a male with one band, which appeared on the result of electrophoresis, ranged from 300-400 base pairs.
