Abstract
The purpose of this research was to know the cross-reaction between antigens of Ascaridia galli with serum anti-Heterakis gallinarum which stated through relative molecular mass. The protein fractions can be used as an advanced research in molecular biology to be the development of candidate vaccines and diagnostic helminthiasis in poultry. Adult stage of A. galli has been collected from the small intestine of domestic poultry, meanwhile H. gallinarum has been found in caecum from Keputran Traditional Market, Surabaya. After that, the worms crushed to obtain whole worm extract (WWE). The Results of WWE has been analized to get protein levels through Bradford’s method, protein levels of A.galli are 3.692 mg/ml and H.gallinarum are 0.546 mg/ml. The results of the analysis of protein levels H._gallinarum can be used as a dose immunization in mice and dose dilution in sample preparation. The analized protein of A. galli and H. gallinarum has been done by using Sodium Deodecyl Suphonat Poly Acrylamide Gel Electrophoresis’s method (SDS-PAGE). Antigens of A.galli used to do cross-reaction with serum anti-H._gallinarum using Western blot’s method. The results showed that cross-reaction protein of A.galli with serum anti-H.gallinarum using Western Blot’s method obtained 10 protein bands with each relative molecular mass are 180 kDa, 148 kDa, 124 kDa, 80 kDa, 54 kDa, 44 kDa, 34 kDa, 31 kDa, 22 kDa and 19 kDa.
