Abstract
The aim of this study was to know the influence of liquid sugar concentration, incubation period, and the best combination towards Bacillus subtilis 3KP biosurfactant production. The study was conducted in completely randomized design with pattern 4x6 and three replications. Growth medium which was used in this study, consist of synthetic mineral water with the addition of liquid sugar in various concentrations (0, 1, 2, and 4%). 4% Bacillus subtilis 3KP starter was inoculated into the medium (OD = 0.5 at A = 610 nm) and incubated on a shaker 120 rpm at room temperature. The biosurfactant production was observed at the various incubation periods (0, 1, 2, 3, 4, and 5 days). The data of surface tension was analyzed using Brown-Forsythe test and continued by Games Howell test. Data of emulsification activity and the weight of biosurfactant crude product were analyzed using Kruskal Wallis test and continued by Mann Whitney test. The result of this study showed that liquid concentration, incubation period, and the combination between both of them influence Bacillus subtilis 3KP biosurfactant production. The best combination of biosurfactant production by Bacillus subtilis 3KP was 4% concentration and 4 days incubation period.
