Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh perbedaan konsentrasi molase, amonium nitrat, dan kombinasi keduanya terhadap jumlah sel dan biomassa bakteri Bifidobacterium bifidum. Konsentrasi molase dan amonium nitrat yang digunakan sebagai media kultur adalah : 0%; 2,5%; 5%; 7,5% dan Og/l, 5g/1, lOg/1, 15 g/l. Starter bakteri B. bifidum sebesar 10% dari media kultur diinokulasikan ke dalam masing-masing media kultur, diinkubasi selama 4 hari, kemudian pada hari ke 4 dilakukan pengukuran pertumbuhan bakteri B. bifidum dengan metode TPC (Total Plate Count) dan pengukuran biomassa. Metode TPC dilakukan dengan cara melakukan seri pengenceran yang representatif kemudian menanam hasil pengenceran tersebut ke dalam media de Man Rogosa and Sharpe (MRS) Agar, dan menghitung koloni yang tumbuh. Metode penghitungan biomassa dilakukan dengan cara mensentrifugasi media kultur, mengambil pelet biomassa yang terbentuk, kemudian menimbang berat kering pelet biomassa. Data yang didapat dari pengukuran pertumbuhan menggunakan dua metode tersebut dianalisa menggunakan ANAVA dua arah dilanjutkan dengan uji Beda Nyata Terkecil (BNT) dengan taraf signifikansi 5% dan diikuti dengan uji Duncan. Hasil pengukuran dengan metode TPC menunjukkan bahwa konsentrasi molase, amonium nitrat, dan kombinasi keduanya berpengaruh nyata terhadap jumlah sel bakteri B. bifidum. Jika diukur dengan metode pengukuran biomassa, konsentrasi molase dan amonium nitrat menunjukkan pengaruh nyata terhadap biomassa bakteri B bifidum, sedangkan kombinasi diantara keduanya tidak menunjukkan pengaruh nyata. Konsentrasi molase yang terbaik adalah 5% (15,57x107 CFU/ml dan 87,93 mg), amonium nitrat yang terbaik adalah 5 g/1 (14,91x107 CFU/ml dan 79,21 mg), dan kombinasi molase dan amonium nitrat yang terbaik adalah pada kombinasi molase 5% dan amonium nitrat 5 g/1 (50x107 CFU/ml dan 107,97mg). Translation: This research was aimed to know the effect of different consentration of molasses, ammonium nitrate, and the combination of both molasses and ammonium nitrate to the growth of Bifidobacterium bifidum bacteria. Molasses and ammonium nitrate consentration used in this research were: 0%, 2,5%, 5%, 7,5% and 0 g/l, 5 WI, 10 WI, 15 g/1. 10% B. bifidum starter was inoculated into each culture media and incubated. At the 4th day of incubation, the measuring of B. bifidum growth using TPC (Total Plate Count) method and biomass counting were done. TPC method were done by doing diluting series, inoculating the representative dilution, and counting the B. bifrdum colony which grew up in the de Man Rogosa and Sharpe (MRS) Agar media. The biomass counting were done by sentrifuging the culture media, taking the pellet biomass, and weighing the dry weight of the pellet biomass. Data gained from the measuring of B. bifidum growth were analyzed by two-way ANOVA and LSD (Least Significant Different) test at 5% level and followed by Duncan test. The result of TPC method showed that the consentration of molasses, ammonium nitrate and the combination of them were influential to the number of B. bifidum cell. The result of counting biomass showed that the consentration of molasses and ammonium nitrate were influential to the biomass of B. bifidum, whereas the combination of them was not influential. The best consentration of molasses was 5% (15,57x107 CFU/ml and 87,93mg), amonium nitrate was 5 g/l (14,91x107 CFU/ml and 79,21 mg), and the combination of molasses and ammonium nitrate were 5% molasses and 5 g/l ammonium nitrate (50x107 CFU/ml and 107,97mg).
