Abstract
Tujuan penelitian ini adalah mengetahui pengaruh ekstrak metanol lichen Ramalina javanica pada konsentrasi yang berbeda terhadap diameter daerah penghambatan (halo) Staphylococcus aureus, Escherichia coli, dan Aspergillus fumigatus dan untuk mengetahui konsentrasi minimum yang dapat menghambat (MIC) dan membunuh (MBC/MFC) mikroba uji tersebut. Konsentrasi yang digunakan pada penelitian ini adalah 0 ppm, 2.000 ppm, 4.000 ppm, 8.000 ppm, 20.000 ppm dan 40.000 ppm. Uji aktivitas antimikroba menggunakan metode cakram kertas (diffusion method) dan metode pengenceran dalam tabung (dilution method). Diameter daerah penghambatan dianalisis dengan uji ANOVA satu arah dengan derajat signifikan 0.05, dilanjutkan dengan uji LSD untuk mengetahui beda nyata terkecil antar perlakuan. Sedangkan, jumlah koloni mikroba dari hasil metode pengenceran dalam tabung (dilution method) dianalisis secara deskriptif untuk mendapatkan nilai MIC dan MBC/MFC. Dari hasil penelitian ini, diketahui bahwa ekstrak metanol lichen R. javanica pada konsentrasi yang berbeda memberikan pengaruh terhadap pertumbuhan S. aureus, sedangkan terhadap E. coli dan A. fumigatus tidak memberikan pengaruh. Ekstrak metanol lichen R. javanica menghambat S. aureus pada konsentrasi 20.000 ppm (MIC) dan membunuh pada konsentrasi 40.000 ppm (MBC). Konsentrasi minimal penghambatan (MIC) dan membunuh (MBC/MFC) dari ekstrak metanol R. javanica terhadap E. coli dan A. fumigatus sampai pada konsentrasi 40.000 ppm tidak didapatkan. Translation: The purpose of this research was to know the influence of methanolic extract of Ramalina javanica lichen to different concentrations on the inhibition area diameter (halo) of Staphylococcus aureus, Escherichia coli, and Aspergillus fumigatus, and to know the inhibiting activity (MIC), and its bacteriocidal/fungicidal (MBC/MFC) to the tested microbes. The used concentrations which were used in this research were : 0 ppm, 2.000 ppm, 4.000 ppm, 8.000 ppm, 20.000 ppm, and 40.000 ppm. The antimicrobial activity test used diffusion method and dilution method. The inhibition area diameter was analyzed by ANOVA one-way test with significance degree: 0,05, continued by LSD test to know the least significance different between the treatments. While, the amount of microbe colonies of the dilution method were analyzed to get MIC and MBC/MFC values. From this research was known that methanolic extract of R. javanica lichen in different concentrations, affected to S. aureus growth. While for E. coli and A. fumigatus methanolic extract of R. javanica lichen in different concentrations did not affect. Methanolic extract of R. javanica lichen inhibited S. aureus at 20.000 ppm (MIC) and its bacteriocidal value was at 40.000 ppm (MBC). The minimal inhibitory concentration of methanolic extract of R. javanica lichen and its bacteriocidal/fungicidal concentration to E. coli and A. fumigatus until 40.000 ppm, there was no value yet.
