Abstract
This research is carried out to analyze molecular order, not only nucleotide, but also protein and paratope structure of Encoding Gene of scFv about heavy chain variable (VH) and light chain variable (VL) of Immunoglobulin G from spleen’s mice after vaccinated by inactive multivalent as Immunotherapy prototype.in control group (KK), mice injected with PBS of dosage 0.3 ml, and in treatment group (KP), mice are injected with inactive multivalent dengue vaccine of dosage 0.3 ml. collected spleen cells and flushing. The suspension is discarded, and then pellet is stored in freezer, is being done of RNA extraction. RT-PCR and PCR is carried out to get DNA target encoding gene scFVIgG, then being seen on electrophoresis gel. Purification of PCR result uses QLAquick Gel Extration Kit. DNA will be readed in sequencer machine, ABI 3130 Genetic analyser. Molecular Analysis of scFv encoding gene is carried out in light chain variable (GLV) and heavy chain variable (GHV). The result of paratopeanalyse is, in control group, it has 6 ABR, and in treatment group, it has 6 ABR.
