Abstract
Objectives: To investigate whether Sargassum duplicatum ethanol extract (SDE) was able to reduce the extent of joint damage through inhibition of NF-B activation in adjuvant arthritic (AA) an animal model of rheumatoid arthritis exposed to cold stressor Material and Methods: Male AA rats (age=10-12 wk; 5 group, n=10/group) were exposed to cold stress (5oC for 15 min/day for 7 days) with/without SDE 400 mg/kg BW/day per oral. First group AA rats did not receive either cold stressor exposure nor SDE. Second group AA rats received only cold stressor exposure. The third group AA rats received SDE starting 7 days prior to cold stressor exposure until the end of cold stressor. The fourth group AA rats received SDE starting 7 days prior to cold stress exposure until 7 days after the end of cold stressor. The fifth group AA rats received SDE starting the first day of cold stressor exposure until 7 days after the end of cold stressor. At the end of the experiment plasma MDAlevelwas measured using spectrophotometry method, joint NF-B levelwas measured by NF-B positive cells in imunohistochemistry (IHC), joint MMP-1 was measured by MMP-1 positive cells in IHC, joint MMP-3 was measured by MMP-3 positive cells in IHC and the severity of joint damage was measured by the histopathological scoring system. Results: Cold stressor exposure in the second group significantly increased plasma MDA level (p≤0.05), increased joint NF-B positive cells (p≤0.05), increased joint MMP-1 positive cells (p≤0.05), increased joint MMP-3 positive cells (p≤0.05) and increased joint histopathological score (p≤0.05) compared to the first group which did not received cold stressor nor SDE. SDE administration in cold stress group 3, 4 and 5 significantly decreased plasma MDA level (p≤0.05), decrease joint NF-B positive cells (p≤0.05), decreased joint MMP-1 positive cells (p≤0.05), decreased joint MMP-3 positive cells and decreased joint histopathological score (p≤0.05) compared to the second group which received cold stressor. Conclusion: Cold stressor significantly increased joint damage and treatment using Sargassum duplicatum ethanol extract 400 mg/kg BW/day per oral significantly reduced joint damage through inhibition of NF-B expression and reduction of stress oxidative in adjuvant arthritic rats.
