Abstract

The research was aimed to explain the mechanism of pup low birth weight of Toxoplasma gondii infected mice by the molecular change of skeletal muscle. The research was randomized true experimental design (posttest only control group design). Thirty two Balb/c mice at twelve weeks old were matched using induction of PMSG and HCG. After sixteen days gestation, to those mice were divided into two groups: treatment and control groups. Treatment used T. gondii RH strain infection at dose of 5 tachyzoites per head. After treatment, those mice were kept until partum. Pups born were counted and balanced for gaining born weight then those pups were sacrificed. Skeletal muscle tissue was performed apoptosis and immunohistochemistry technique observations to know the presence of tissue macrophage of skeletal muscle that expressed TNF- , MyoD and skeletal muscle expressed MMP-9. In order to test hypothesis, data analyzed using t test. The association among variables analyzed with Pearson correlation test. The results showed the presence of increasing tissue macrophage number of skeletal muscle that expressed TNF- , increasing skeleton muscle cell apoptosis and skeletal muscle expressed MMP-9 and decreasing skeleton muscle cell expressed MyoD in born pup of T. gondii infected mice. There was significantly differences (P<0.01) between tissue macrophage of skeletal muscle that expressed TNF- and skeletal muscle expressed MMP-9 of born pup of T. gondii infected and uninfected mice. There was no significantly differences (P>0.05) expression of MyoD and apoptosis in skeletal muscle of born pup of T. gondii infected and uninfected mice. There was relationship between increasing apoptosis with decreasing expression of MyoD in skeletal muscle of born pup of T. gondii infected mice. In conclusion that infection of T. gondii in gestation mice could cause the molecular change of TNF- expression by tissue macrophage skeletal muscle and expression of MMP-9 and those variables play role in the mechanism of low birth weight.
