Abstract

Background: AIDS epidemic has spread to all parts of Indonesia and currently more than 150 countries reported the existence of HIV/AIDS from around the world. Additionally, HIV/AIDS treatment using ARV drugs also find obstacles that must be faced in terms of host, environment and the agent. The objective of this study was to generate lymphocytes TCD4+ that are resistant to HIV infection through transfection of CCR5 mutant 32 bp encoding gene to bone marrowhematopoietic stem cell. Method: This type of study was an in-vitro study with factorial research design. The 4 groups were subjected to 32 bp nucleotide deletion of the CCR5 encoding gene, cloned and sub cloned, and than transfected by chemical methods with or without the addition of cloroquin to bone marrow haematopoietic stem cells. To check resistance of HIV infection was tested by co-cultivation by PBMC of HIV patients, and formation of FFU (Focus Forming Units) were observed. Result: The results of statistical analysis by using multiway ANOVA showed that the main effect of type of DNA that was fully transfected CCR5 gene as compared with CCR5 mutant 32 bp had the highest significance level of p = 0.000. Thus, this study found that the finally FFU was formed very low on the CCR5 mutant 32, it indicated high resistance to HIV infection. Conclusion: Transfection mutant 32 bp CCR5 gen coding toBone Marrow hematopoieticStem Cell causes lymphocyte TCD4+ cells become resistant to HIV infection. On the future this product will be used to treat for HIV/AIDS cases
