Abstract
Iron-deficient anemia remains a remarkable nutritional problem in Indonesia. It is recognized that ascorbic acid is one of iron-absorption enhancer factor, and citric acid is an organic compaund that acts synergistically with ascorbic acid in enhancing iron absorption. The aim of this study was to find out whether the administration of ferrous sulfate, ascorbic acid, and cirtic acid either alone or in combination, with converted dose may increase iron status in iron-deficient rats. Sixty female Wistar strain rats of 4 weeks old were used.The Post-test Only Control Group Design was used to analyze the data in this study. Before treatment, those rats were rendered to be iron-deficient by giving low-iron diet for 8 weeks. After becoming iron-deficient, the animals were divided into 4 treatment groups, as follows : TI = ferrous sulfate 3.6 mg/day; T2 = ferrous sulfate 3.6 mg/day and ascorbic acid 4.5 mg/day; T3 = ferrous sulfate 3.6 mg/day and citric acid 5.4 mg/day; T4 = ferrous sulfate 3.6 mg/day, ascorbic acid 4.5 mg/day and citric acid 5.4 mg/day. The dose was converted dose with correction factor of 0.018. Parameters used to measure iron status in rats were Hb level, serum iron (SI), total iron binding capacity (TIBC), and transferrin saturation percentage. Results showed that after being treated for 2 weeks, all groups experienced increased iron status, indicated by the increase of lib level, SI and transferrin saturation percentage, and decreased TIBC level significantly (p<0.05) compared to that before treatment (during iron-deficient condition). Hb level increase from 11.3 gram % to 15.02 gram %, SI level increase from 72.29 ug/dl to 223.28 ug/dl, TIBC level decrease from 543.15 ug//dl to 479.627 ug/dl and transferrin saturation increase from 13.32% to 46.61 %. From all treatment group, P4, which treated with combined ferrous sulfate, ascorbic acid, and citric acid, showed the highest increase of iron status compared to other treatment groups.
